Culture Media & Dehydrated Media: Complete Guide for Microbiology Labs Pakistan

Nutrient agar, MacConkey, blood agar base, LB broth, dehydrated culture media — preparation, storage, and sourcing for Pakistan labs.

What Is Dehydrated Culture Media?

Dehydrated (powder) culture media is the standard format for microbiology labs in hospitals, food factories, universities, and pharma. You reconstitute with distilled water, sterilize by autoclave, and pour plates or prepare broths.

Advantages of Dehydrated vs Ready-Made Plates

Powder media costs less per test, ships without refrigeration, and offers longer shelf life when stored dry. High-volume food and water testing labs in Pakistan typically prepare plates in-house.

Essential Dehydrated Media List

  • General purpose: Nutrient agar, nutrient broth, tryptic soy agar (TSA), tryptic soy broth (TSB)
  • Selective/differential: MacConkey agar, EMB agar, XLD agar, SS agar, Baird-Parker agar
  • Blood media base: Blood agar base (add defibrinated sheep blood 5–7%)
  • Enrichment: Selenite broth, alkaline peptone water, Rappaport-Vassiliadis (RV) broth
  • Molecular biology: LB agar, LB broth, Miller and Lennox formulations
  • Fungi: Sabouraud dextrose agar (SDA), potato dextrose agar (PDA)
  • Water testing: m-Endo, m-FC, R2A agar

Preparation Tips

  • Weigh powder on analytical balance — do not estimate
  • Adjust pH before autoclaving if specified
  • Autoclave at 121°C for 15 min (verify with indicator tape)
  • Cool agar to 50°C before adding heat-sensitive supplements
